Sequence Verification Help File |
|
The 7409 cDNA clones of the NIA Mouse 7.4K cDNA Clone Set are arrayed among 79 96-well microtiter plates (numbered H4001 to H4079).
Each Sequence verification page is a diagram depicting, in the format of a 96-well microtiter plate, all of the sequence verification information. That is, each well of each plate is represented by the row and column heading on the page. Please visit http://lgsun.grc.nia.nih.gov/help.html for details regarding clone addressing/naming.
Each cell in the diagram has a number or letter(s). The number represents the "Score" (see 15K help file) of the homology between the 3' sequences of the 15K rearray and each of their source (Parent) clone 3' sequence. The letter "U" for Unverifiable, indicates that the sequence information obtained from resequencing is not sufficient to verify either the sequence or the clone's residence. The letter "O" for Orphan, indicates that the sequence has been verified by multiple sequence attempts, but that the sequence indicates no definitive connection between the rearrayed clone (Daughter) and it's original source (Parent) clone. In most cases it is clear that there is a problem with the addressing of the source (Parent) clones, such as two plates having been switched for example. Since these instances have yet to be resolved, but the sequences have been verified by further sequencing, we are calling these clones "verified" but we have termed them "Orphans." Efforts are being made to resolve the few instances where this occurs.
Either of the 3' or 5' sequence information for each is sufficient to verify the clone's residence. For Orphan Clones if the clone is verified by using 5' sequence information then you will see the clone column in light blue color otherwise you will see another color.
CAUTION: The sequence verification provided here should never be taken as a substitute for sequencing clones isolated for further experiments.
